IL DIRETTORE GENERALE
SU PROPOSTA dell'U.O. Gestione Ricerche e Progetti Speciali;
PREMESSO che l'Istituto Zooprofilaltico Sperimentale dell'Umbria e delle Marche annovera tra le
finalità l'attività di ricerca e che la stessa tra le voci di spesa annovera quella per missioni, che
deve intendersi a spese riferibili a partecipazione a convegni.
VISTO il modulo di autorizzazione per la partecipazione a convegni/corsi/acquisizione tecniche
innovative e missioni del personale dell'Istituto per attività di ricerca con il quale:
1. Chiara Magistrali, Veterinario Dirigente, richiede l'autorizzazione per la partecipazione al
6'h European Symposium of Porcine Health Management che si terrà a Sorrento dal 7 all'8
maggio 2014, da imputare alla RC0132012;
2. Monica Cagiola, Veterinario Dirigente, richiede l'autorizzazione per la partecipazione al
12'h International Colloquium on Paratuberculosis, che si terrà Parma dal 22 al 26 giugno
2014, da imputare alla RFPA12012;
VISTA la delibera 57/2014 con la quale è stata autorizzata Roberta Orletti alla partecipazione al
5th International IUPAC Symposium
for Trace Elements in Food (TEF-5) Copenaghen
(Danimarca), dal 6 al 9 maggio 2014; visto altresi che la fattura emessa comprende anche le
spese relative all'albergo e che pertanto la spesa complessiva da liquidare in fattura è
rideterminata in €. 957,77 restando ferma l'imputazione alla RC0052011;
VISTO l'art. 6, comma 12, del D.L. 78/2010, a norma del quale a decorrere dall'anno 2011 le
amministrazioni
pubbliche
inserite nel conto economico
consolidato
della
pubblica
amministrazione, non possono effettuare spese per missioni, anche all'estero, per un ammontare
superiore al 50 per cento della spesa sostenuta nell'anno 2009, con esclusione di quelle
indispensabili per assicurare /a partecipazione a riunioni presso enti e organismi internazionali o
comunitari;
DATO ATTO che le missioni in argomento rientrano nella fattispecie esclusa dal limite di spesa ai
sensi dell'art. 6 citato, in quanto le partecipazioni sono legate a progetti di ricerca con finanziamenti
definiti nel progetto per la partecipazione a convegni e/o riunioni;
VISTA la deliberazione del Consiglio di Amministrazione n, 10 del 18.09,2002 con cui è stato
approvato il Regolamento per l'espletamento delle missioni;
VISTO inoltre l'art, 8 "Trattamento economico di missione all'estero - Personale di tulte le
categorie", e le successive modificazioni intervenute con Decreto Legge n, 78 del 31/05/2010
convertito con Legge n, 122 del 30/07/2010;
VISTE le deliberazione del Direttore Generale n, 106 del 13,05.2011, n. 257 del 14.10,2011 e n, 9
del 18.01,2012 con cui sono stati definiti i budget per le missioni e per i costi relativi ai progetti di
ricerca, ai sensi delle quali:

risorse e che l'U O. Controllo di Gestione gestisce come centro di costo di attività, le
autorizzazioni e le responsabilità sono di esclusiva competenza del Responsabile Scientifico
della ricerca stessa e del Responsabile Amministrativo dell'Ufficio Ricerche;
VISTA la deliberazione del Direttore Generale n. 106/2011 che definisce il conto n. 52221312 per
spese relative alle quote di iscrizione ai Convegni in territorio nazionale ed estero;
VISTE le richieste di partecipazione a convegni pervenute all'U.O. Gestione Ricerche e Progetti
Speciali, formulate dagli interessati nell'apposito modulo RP101 per il personale a tempo
indeterminato, allegate alla presente per farne parte integrante e sostanziale;
DATO ATTO, pertanto, che l'U.O. Gestione Ricerche e Progetti Speciali, dal 0610612011,
provvederà ad inoltrare le iscrizioni del personale per la partecipazione a Convegni indicate nella
tabella n. 1 allegata;
RILEVATO che il presente provvedimento é autorizzativo dei costi di iscrizione ai Convegni come
da tabella n. 1, e dei relativi costi di missioni;
RILEVATO che i costi complessivi di iscrizione e missione per la partecipazione del personale alle
attività formative indicate nell'allegato tabella n. 1, rientrano nei budget assegnati nei vari progetti
di ricerca alle varie Articolazioni OrganizzativelResponsabili Scientifici;
VERIFICATA la disponibilità residua del piano finanziario delle ricerche da parte dei Responsabile
Scientifici delle stesse;

Abstract text:
Traditional diagnosis of Paratuberculosis (PTB) is based on serology and fecal culture. Gamma
Interferon (y-IFN) and Skin Test (5T), already used far ante-mortem diagnosis of bovine Tuberculosis
(bTB) could be also useful for PTB diagnosis. y-IFN test detect.s cytokine production of T Iymphocytes
after stimulation with purified protein derivatives (PPDs), fram M. bovis (PPDS) and from M. avium
(PPDA). In arder to achieve an earty detection of subjeds exposed to MAP infect.ion, 3 new PPDs
extracted from culture of MA? (PPDJ a, b, c) were produced, assessed in the y-IFN test, compared to
c1assic PPDs. In the ongoing study, 68 caUle from bTS officially free herds and with previous PTB
c1inical cases were tested by IDVet ELISA, PCR and culture. PPDJs were used in y.IFN test
(BOVIGAM ELISA) al dilutions 1:5 and 1:10. Samples have been considered positive when Optical
Density (OD) value was, at least, twice OD of Nihil. Relative specificity (Sp) of PPOS and PPDJ in
bTB diagnosis was calculated using 5T as Gold Slandard (G5). Relative sensitivity (Se) of PPDJ in
PTB diagnasis was calculated using, as G5, the positivity in at least one of c1assical tests and peR.
aut of 55 animaJs PTB positive, 46 reacted to both PPDA and PPOJ, 4 only to PPDJ. For bTS
diagnosis, the comparative use of PPDA and PPDJ against PPDB in 68 bTB negative cows yielded a
100% Sp. For PTB diagnosis, 1:5 dilution of PPDJ a, band c showed respectively 83.6%, 76.4% and
74.5% Se. Relatively low Se in PTB diagnosis is influenced by tests used as GS, which are able to
detect only advanced stages of disease. In fad 4 aut of 9 subjects, previously c1assified as PTB
negative, but positive to PPDJ, became positive 8 months laler to serological and culture tests. Our
preliminary results highlight the ability of our y-IFN to avoid false positivity for bTB. For PTS diagnosis
we pian to include more subjects with a suitable follow up. Projects RCIZ5UM 11/2008 and RCIZSUM

In many European countries, B. hyodysenteriae, B. pilosicoli and L. intracellularis, are considered
the major causes ofintestinal diseases of growers and fatteners. The symptoms ofthese diseases are
often similar in field conditions and mixed infections are very frequent in field cases, making a
differential diagnosis cumbersome. Multiplex real time PCR , targeting these three pathogens
simultaneously, has shown good performances when compared to traditional tests, and could be
useful when a mixed infection is suspected, to reduce time and cost of a differential diagnosis. Since
this test can not onIy detect, but also quantify the bacteria, it has been suggested that it can be used
to interpret the test outcomes. An association between faecal loads and low growth rate has been
shown for L. intracellularis For B. hyodysenteriae, an inverse relationship between faecal
consistency and faecalloads was suspected in cases of enteritis occurring in the field.
Aim of this work was to confirm the hypothesis of an inverse relationship between the presence of
diarrhoea, the fuecal consistency and the amount of B. hyodysenteriae excreted by artificially
challenged pigs.
One hundred and fifty faecal samples were taken from pigs artificially challenged by B.
hyodysenteriae
21,26,28,34,41,48
days before. The pigs were from Salmonella and B.
hyodysenteriae free herds and tested negative for these pathogens before challenge. The samples
were categorized as non diarrhoeic or diarrhoeic by the same veterinarian, cultured for Brachyspira
spp, and exarnined by real time PCR targeting L. intracellularis and B. pilosicoli. The dry matter
content of sixty seven faeces was evaluated by a microwave procedure; then the faeces were
divided in four different categories, I to 4, with decreasing dry matter content, as aIready described
O.
AlI samples tested negative for L. intracellularis and B. pilosicoli. Non diarrhoeic faeces were
grouped in category one onIy. The baeterialload, expressed as 10glO,increased when diarrhoea was
present (p=O,7011; p-value:SO,05) and it was on average 4,3 log in normal faeces, and 7,1 log in
diarrhoeic samples. An inverse relationship between the dry matter content and the B.
hyodysenteriae
excretion, expressed as 10glO, was shown (p=-0,4983; p-value::::0,05). This
relationship was stronger in diarrhoeic samples (p=-O,8047). The hacterialload in faeces increased
progressively with the dry matter content category (p=O,6734, p-value::::0,05), as it was greater in
faeces belonging to category 3 and 4.
In conclusion, this work confirrns the correlation between B. hyodysenteriae loads excreted by
faeces and the presence and the severity of the diarrhoea, suggesting a possible use of DNA
quantification in interpreting test results.
